Category:Mughal Empire SufisProstaglandin E2 (PGE2) and its metabolite, 2-arachidonoyl glycerol, are involved in both the initiation and progression of human colorectal carcinogenesis. This proposal builds on recent work showing that PGE2 receptor signaling is required for the ability of the intestinal epithelium to regulate the outgrowth of intestinal tumors. An interaction between the PGE2-cAMP-CREB and phosphoinositol 3-kinase-Akt1 pathways regulates epithelial proliferation and suppresses tumorigenesis. PGE2 also induces a discrete Ca2+ wave in intestinal epithelial cells that propagates to neighboring cells, which leads to a G-protein dependent activation of the ERK pathway and a subsequent induction of genes encoding anti-apoptotic proteins. We hypothesize that the three pathways are activated in concert to control the balance between cell survival and cell death in intestinal epithelial cells and that the relative activation of the pathways determines whether a cell proliferates or dies. We will characterize the interaction of the pathways with the PGE2 receptors in the normal human epithelium and in colorectal tumors. We will identify other intracellular proteins that interact with the pathways in colonic epithelial cells and characterize the activation of the pathways in colon cancer cell lines and patient-derived tumor samples. The role of the pathways in the tumorigenic phenotype of the cells will be assessed by comparison of the signaling pathways in cells with different abilities to induce tumors in animals. We will identify the transcription factors that govern cell survival and cell death, and define their role in the survival of cells that fail to undergo apoptosis in vivo.
